Right click on "My Computer" and go to properties, go to hardware tab, device manager, sound & video (or similar), find your Audio driver, disable, click ok, then re-enable, click ok. If this doesn't work try updating the driver anyway. If this doesn't work uninstall then re-install the driver. If you have the product recovery disk that came with your computer all the drivers will be on it, if not you can most likely find the driver or updated driver on your computer manufacturers website or the audio card manufacturers website. It could be possible that your audio card is toast, but go after software problems first. It's probably something simple like a corrupt driver or for some reason the service isn't being started when your operating system starts up.

In the hardware/device manager - sometimes the easiest thing to do is delete the sound all together - then just reboot or run the "Add Hardware" wizard from the control panel. Just my 2 cents...if it's broke, it won't see it and you'll know then you need to go pick up a sound card.
